Project summary :
In the CONECT-AML project, we are developing models for studying mutations associated with aggressive subgroups, either directly from patient cells (PDX: patient-derived xenograft), or by expressing these mutations in normal cells . We have developed models from cells of leukemia patients (Thiollier et al. JEM 2012, Thirant et al. Cancer Cell 2017) and a mouse model showing certain bases of the pediatric specificity of these leukemias (Lopez et al. Cancer Discovery 2019). We are currently moving towards the development of models from normal human cells, including from induced pluripotent cells, in order to understand the trajectory of the transformation from a normal cell to a leukemic cell. These models also allow molecular analyzes (eg epigenetic and gene expression regulations) with the aim of highlighting mechanisms presenting new therapeutic perspectives (Aid et al. Leukemia 2022).
